What's the difference between dihydroberberine and berberine?

Berberine is a plant alkaloid. Dihydroberberine (DHB) is its reduced form — the same molecule, with two extra hydrogen atoms, that your gut microbiome naturally turns berberine into so it can cross the intestinal wall. DHB is absorbed far more efficiently than berberine, then oxidizes back to berberine once it's in your tissues. In other words, DHB isn't a different or synthetic compound — it's berberine's own absorbable intermediate. Supplementing it directly skips the bottleneck your body would otherwise have to work around.

What is berberine?

Berberine is a bright-yellow alkaloid found in plants like goldenseal, barberry, and Oregon grape, used in traditional medicine for centuries. Today it's one of the more heavily studied botanicals for metabolic and healthy-glucose-metabolism support. Most of that research pairs it with the same practical problem, though: getting it into the body.

Berberine's catch: your body barely absorbs it

Standard berberine has famously poor oral bioavailability — under 1%, with one careful analysis putting the absolute figure near 0.68%.¹ Between low solubility, a cellular "pump" (P-glycoprotein) that ejects it back into the gut, and rapid first-pass metabolism, most of a berberine dose never makes it into circulation. (This is true of berberine HCl too — the form in most "berberine" supplements. The hydrochloride salt changes how the powder dissolves, not the underlying absorption problem.) That's why typical protocols call for 1,000–1,500 mg a day, split into two or three doses — a lot of capsules — and why higher doses are where the familiar complaints of cramping and loose stools tend to show up.

Where DHB comes from — your gut already makes it

Here's the elegant part. Researchers found that gut bacteria convert berberine into dihydroberberine using enzymes called nitroreductases, and that DHB is absorbed at roughly five times the rate of berberine in the intestine before converting back to berberine in the body.² This is the natural pathway that lets any berberine work at all. Taking dihydroberberine directly simply hands your body the form it was going to have to make anyway — which is also where the "5×" in our product name, Berberine⁵, comes from.

The absorption data: DHB vs. berberine in humans

The clearest human comparison is a randomized, controlled, crossover trial that measured berberine in the blood after each form. Over two hours, 100 mg of dihydroberberine reached about 6.7× the blood berberine of a 500 mg dose of standard berberine — a fifth of the dose, several times the exposure.³

Fewer capsules, gentler on the gut

Because a small amount of DHB delivers what a large amount of berberine can't, two practical things change:

  • Lower pill burden. Roughly 100 mg of DHB is equivalent to about 500 mg of standard berberine by blood exposure — so you're not swallowing a fistful of capsules to reach a meaningful amount.
  • Better tolerance. Much of berberine's GI reputation comes from the large, unabsorbed fraction sitting in the gut at high doses. Delivering a smaller, better-absorbed dose sidesteps a lot of that.

  • Check the milligrams: an effective DHB serving is usually in the 100–200 mg range, not the 500–1,000 mg you'd see on a berberine label.
  • Be wary of products that list only "berberine" at a low dose and imply premium absorption without a form that delivers it.
